目的探讨埃兹蛋白(Ezrin)和第10号染色体同源缺失性磷酸酶-张力蛋白基因(PTEN)蛋白在胃癌组织芯片中的表达。方法手术后病理诊断明确的485例原发性胃癌患者,术前均未经放、化疗。其中,高、中分化胃癌254例,低分化胃癌231例,有淋巴结转移者353例,TNMⅠ、Ⅱ期166例,Ⅲ、Ⅳ期319例;另外取距肿瘤7cm的正常胃黏膜组织40例。制成8个组织芯片蜡块,用免疫组化EnVision法检测石蜡包埋的胃及胃癌组织中的Ezrin和核PTEN蛋白表达。结果与正常胃黏膜组织相比,Ezrin蛋白在胃癌组织中高表达,并与肿瘤TNM分期相关(P<0.05);而胃癌组织中核PTEN蛋白呈低表达;两者均与肿瘤分化程度、淋巴结转移相关(P<0.05);Ezrin同时还和核PTEN蛋白表达呈负相关(r=-0.106,P<0.05)。结论 Ezrin可能是预测胃癌组织转移的有用的指标;联合检测Ezrin和核PTEN蛋白可作为判断胃癌预后和筛选高危胃癌转移患者的有效指标。
Objective To investigate the expression of Ezrin and PTEN protein in gastric cancer tissue microarray. Methods 485 patients with pathologically diagnosed primary gastric cancer after surgery were preoperative radiotherapy and chemotherapy. Among them, there were 254 cases of high and moderately differentiated gastric cancer, 231 cases of poorly differentiated gastric cancer, 353 cases of lymph node metastasis, 166 cases of TNMⅠand Ⅱ, 319 cases of stage Ⅲ and Ⅳ, and 40 cases of normal gastric mucosa from 7cm of tumor. Eight tissue chip wax blocks were made and the expression of Ezrin and nuclear PTEN protein in paraffin-embedded gastric and gastric cancer tissues was detected by EnVision immunohistochemistry. Results Compared with normal gastric mucosa, Ezrin protein was highly expressed in gastric cancer tissues and correlated with tumor TNM stages (P <0.05), while nuclear PTEN protein was low expressed in gastric cancer tissues. Both of them were correlated with tumor differentiation and lymph node metastasis (P <0.05). Ezrin also negatively correlated with nuclear PTEN protein expression (r = -0.106, P <0.05). Conclusion Ezrin may be a useful predictor of gastric cancer metastasis. Combined detection of Ezrin and nuclear PTEN protein may be an effective marker for the prognosis of gastric cancer and screening of patients with high-risk gastric cancer.
