目的研究深圳市公共场所冷却塔水中嗜肺军团菌的基因特征。方法采用军团菌巨噬细胞感染力增强因子基因(Macrophage Infectivity Potentiator,mip)分型方法。提取深圳市51株嗜肺军团菌菌株DNA,聚合酶链式反应(Polymerase chain reaction,PCR)技术扩增mip基因,并将PCR扩增产物进行测序。结果每菌株测序后,序列与European Working Group for Legionella Infections(E.W.G.L.I)组织建立的数据库进行比对。51株嗜肺军团菌分为5种型别,分别为L.pnuemophile-phil-1、L.pnuemophile-sg3、L.pnuemophile-D-15、L.pnuemophile-sg10与L.pnuemophile-97-2898。结论mip分型方法可用于嗜肺军团菌的基因分型。深圳各区不同场所冷却塔水中分离的嗜肺军团菌株之间存在差异,L.pnuemophile-phil-1型别为优势型别占60.8%。
Objective To study the genetic characteristics of Legionella pneumophila in the cooling towers of public places in Shenzhen City. Methods The Macrophage Infectivity Potentiator (mip) typing method was used. The DNA of 51 Legionella pneumophila strains in Shenzhen was extracted and the mip gene was amplified by Polymerase chain reaction (PCR), and the PCR products were sequenced. Results After sequencing of each strain, the sequences were aligned with databases established by the European Working Group for Legionella Infections (E.W.G.L.I). 51 strains of Legionella pneumophila were divided into 5 types: L. pnuemophile-phil-1, L. pnuemophile-sg3, L. pnuemophile-D-15, L. pnuemophile-sg10 and L. pnuemophile-97-2898 . Conclusion The mip typing method can be used for genotyping Legionella pneumophila. There was a difference between the Legionella pneumophila strains isolated from cooling tower water in different places in Shenzhen, and L.pnuemophile-phil-1 type accounted for 60.8% of the dominant type.
