目的:观察当归挥发油经婴儿皮肤的渗透动力学特性,并从形态学的角度探讨当归挥发油促进药物透皮吸收的机制。方法:采用Franz扩散池进行体外渗透实验,HPLC法检测接收液中藁苯内酯的浓度,计算当归挥发油的经皮累积渗透量和速率常数。并用扫描电镜和透射电镜观察当归挥发油对皮肤超微结构的影响。结果:不同浓度当归挥发油在婴儿皮肤的渗透行为均符合Higuchi方程;扫描电镜观察显示,当归挥发油使皮肤表面皱褶增多,角质层局部断裂脱屑,翻卷呈破棉絮状,毛囊口扩展,毛干的毛小皮剥脱变细;透射电镜显示,角质层细胞崩解脱落,基底层及棘层细胞连接断裂,细胞间隙增大。结论:当归挥发油具有较好的皮肤渗透性能,其促进药物透皮吸收的机制与改变皮肤结构密切相关,当归挥发油可改变角质层结构,使细胞间隙增大,有利于药物渗透。
OBJECTIVE: To observe the osmotic kinetics of volatile oil from Angelica sinensis on infantile skin and to explore the mechanism of Angelica sinensis volatile oil promoting transdermal absorption of the drug from the morphological point of view. Methods: Franz diffusion cells were used for in vitro permeation test. HPLC was used to determine the concentration of ndaprolactone in the receiving solution. The percutaneous cumulative permeation and rate constants of the volatile oil were calculated. The effects of Angelica sinensis volatile oil on the ultrastructure of the skin were observed by scanning electron microscopy and transmission electron microscopy. Results: The osmotic behavior of Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis Angelica sinensis under various concentrations was in accordance with Higuchi equation. Scanning electron microscopy showed that Angelica sinensis volatile oil increased the surface folds, cut off the sclerenchyma, Of hairy skin peel thinning; transmission electron microscopy showed stratum corneum cells disintegration shedding, basal and stratum spinosum cells connected and broken, cell gap increased. Conclusion: Angelica sinensis essential oil has better skin permeability, and its mechanism of promoting drug transdermal absorption is closely related to the change of skin structure. Angelica sinensis volatile oil can change the structure of the stratum corneum and increase the cell gap, which is good for drug penetration.
