CpG寡核苷酸促进Toll样受体9的表达增强乳腺癌细胞侵袭

目的:探讨Toll样受体9(Toll-like receptor 9,TLR9)在乳腺癌细胞中的表达对其侵袭能力的影响。方法:应用West-ern blot检测乳腺癌细胞系TLR9的表达状况,以CpG寡核苷酸刺激不同乳腺癌细胞株后,检测TLR9表达的变化、体外侵袭能力的变化以及细胞活性的影响。结果:人乳腺癌细胞株MDA-MB-231表达TLR9而MCF-7不表达。CpG寡核苷酸处理后MDA-MB-231的TLR9表达增高,且侵袭能力进一步增强;MCF-7的该受体表达情况和侵袭能力均无明显变化。侵袭能力的增加不是源自于细胞活性的增强。结论:TLR9在高侵袭的乳腺癌细胞系中表达,其表达的增加可以促进乳腺癌细胞的侵袭能力。 Objective: To investigate the effect of Toll-like receptor 9 (TLR9) expression on the invasiveness of breast cancer cells. Methods: The expression of TLR9 in breast cancer cell line was detected by West-ern blot. The expression of TLR9 in different breast cancer cell lines was stimulated by CpG-ODN. The invasion ability and cell viability were detected. Results: Human breast cancer cell line MDA-MB-231 expressed TLR9 but not MCF-7. The expression of TLR9 in MDA-MB-231 cells was increased after CpG oligodeoxynucleotide treatment, and the invasion ability was further enhanced. The expression and invasion capacity of MCF-7 cells did not change significantly. Increased invasiveness does not result from increased cellular activity. CONCLUSION: TLR9 is expressed in highly invasive breast cancer cell lines. The increased expression of TLR9 may promote the invasiveness of breast cancer cells.
