本文采用硷化乙醇法制备细菌O抗原,用以致敏醛化红细胞。临用前加入3%聚乙二醇(PEG)、灭活的正常兔血清及绵羊红细胞膜配成间接血凝应用液,以检测杂交瘤产生的抗霍乱弧菌菌体抗原的单克隆抗体。在细胞培养上清和小鼠腹水抗体的检测中,其灵敏度比经典的醛化致敏血球提高4~8倍。
In this paper, alkaline ethanol method for the preparation of bacterial O antigen, used to sensitize the aldehyde red blood cells. Before use, 3% polyethylene glycol (PEG), inactivated normal rabbit serum and sheep erythrocyte membrane were added to form an indirect hemagglutination solution to detect the monoclonal antibodies against Vibrio cholerae bacteria antigens produced by the hybridomas. In the cell culture supernatant and mouse ascites antibody detection, the sensitivity of the aldosterone-sensitive than the classic increase of 4 to 8 times.
