目的:探讨不同分期的多发性骨髓瘤(MM)患者外周血淋巴细胞各亚群细胞比例与白细胞介素-6(IL-6)表达变化。方法:用流式细胞仪检测35例初治MM患者及10例对照者的外周血淋巴细胞亚群各细胞表达水平,用ELISA法检测IL-6水平。结果:与对照者相比,MM患者的CD4+、CD4+/CD8+明显降低(P<0.01),均与MM分期严重程度呈负相关(r=-0.780,-0.832,P<0.01);MM患者的CD8+显著增加(P<0.01),与MM分期严重程度呈正相关(r=0.737,P<0.01)。MM患者的IL-6水平较对照者显著增加,与MM分期严重程度呈正相关(r=0.961,P<0.01)。结论:IL-6表达增加及T淋巴细胞亚群数目异常变化可能是MM患者免疫异常的机制之一,两者能作为观察MM患者病情严重程度的指标。
Objective: To investigate the changes of the proportion of peripheral blood lymphocyte subsets and the expression of interleukin-6 (IL-6) in patients with multiple myeloma (MM) at different stages. Methods: The expression of IL-6 in peripheral blood lymphocyte subsets of 35 newly diagnosed MM patients and 10 controls were detected by flow cytometry. Results: Compared with controls, MM patients had significantly lower CD4 +, CD4 + / CD8 + (all P <0.01), and were negatively correlated with the severity of MM stage (r = -0.780, -0.832, CD8 + increased significantly (P <0.01), which was positively correlated with the severity of MM stage (r = 0.737, P <0.01). The level of IL-6 in patients with MM was significantly higher than that in controls, which was positively correlated with the severity of MM (r = 0.961, P <0.01). CONCLUSION: Increased expression of IL-6 and abnormal changes in the number of T-lymphocyte subsets may be one of the mechanisms of immune dysfunction in MM patients, both of which can be used as indicators to observe the severity of MM patients.
