目的:探讨治疗银屑病的临床有效外用药芩柏软膏中黄芩的主要成分黄芩苷(baicalin)、黄芩素(baicalein)和汉黄芩素(wogonic)对T细胞早期活化标志CD69表达的影响。方法:以体外培养的Jurkat T细胞为模型,以二丁基佛波醇酯(phorbol 12,13-dibutyrate,PDB)和离子霉素(Ionomycin,Ion)为刺激剂,采用单荧光染色流式细胞技术,检测Jurkat T细胞经PDB+Ion刺激4 h后早期活化表面分子CD69的表达,以观察黄芩苷、黄芩素和汉黄芩素对Jurkat T细胞体外活化的影响。结果:在体外培养条件下,黄芩苷(200,100,50 mg·L-1)、黄芩素(54,27,13.5 mg·L-1)和汉黄芩素(8,4 mg·L-1)均能抑制PDB+Ion刺激时Jurkat T细胞CD69的表达,且有剂量依赖性。结论:黄芩主要成分黄芩苷、黄芩素和汉黄芩素可明显抑制Jurkat T细胞的体外活化,这可能是黄芩用于治疗银屑病的作用机制之一。
Objective: To investigate the effects of baicalin, baicalein and wogonic, the main components of Astragalus membranaceus in the treatment of psoriasis, on the expression of early activation marker CD69 on T cells. METHODS: The Jurkat T cells cultured in vitro were used as the model, with the phorbol 12, 13-dibutyrate (PDB) and ionomycin (Ionomycin, Ion) as stimulants, and the single-fluorescein staining flow cytometry was used. The technique was used to detect the expression of CD69 on surface of Jurkat T cells stimulated by PDB+Ion for 4 h, and to observe the effects of baicalin, baicalein and wogonin on the in vitro activation of Jurkat T cells. Results: Baicalin (200,100,50 mg·L-1), baicalein (54,27,13.5 mg·L-1) and wogonin (8,4 mg·L-1) were all cultured in vitro. The expression of CD69 in Jurkat T cells was inhibited by PDB+Ion stimulation in a dose-dependent manner. CONCLUSION: Baicalin, baicalein, and wogonin can significantly inhibit the activation of Jurkat T cells in vitro. This may be one of the mechanisms of Astragalus in the treatment of psoriasis.
