目的探讨IL-10对支气管哮喘(哮喘)小鼠肺部变应性炎症的抑制作用及对血清IgE水平的影响。方法 30只6~8周龄清洁级雄性BALB/c小鼠,体质量(20±2)g,随机分为正常组,哮喘组和IL-10组,每组各10只。哮喘组和IL-10组以1g.L-1卵白蛋白(VOA)腹腔注射致敏,以10g.L-1VOA20mL雾化吸入激发制作小鼠哮喘模型。正常组不做任何处理。哮喘组每次激发前予9g.L-1盐水0.1mL鼻内滴入,IL-10组每次激发前给予含IL-10(0.2μg.L-1)的9g.L-1盐水0.1mL鼻内滴入。末次激发24h后,经小鼠眼球采血分离血清,采用ELISA法测定其血清IgE水平。取其肺组织行病理切片。结果哮喘组小鼠病理改变与其他各组有明显区别,支气管和血管周围炎性细胞浸润(主要为嗜酸性粒细胞,中性粒细胞),黏膜及黏膜下层水肿明显,呼吸道中可见较多的黏液和黏液栓。而IL-10组与正常组无明显区别:肺泡结构清晰,肺泡腔和支气管腔无渗出物,与哮喘组比较有显著性差异。正常组、哮喘组和IL-10组小鼠血清IgE水平分别为(5.219±0.427)μg.L-1、(6.633±0.145)μg.L-1和(5.503±0.566)μg.L-1,哮喘组血清IgE水平与正常组及IL-10组比较差异均有统计学意义(Pa<0.05),IL-10组与正常组比较差异无统计学意义(P>0.05)。结论 IL-10局部滴入治疗可抑制哮喘小鼠肺部变应性炎症反应,改善其呼吸道症状,降低哮喘小鼠血清IgE水平。
Objective To investigate the inhibitory effect of interleukin-10 (IL-10) on allergic inflammation in the lungs of asthmatic mice and its effect on serum IgE levels. Methods Thirty male BALB / c mice of 6-8 weeks old were randomly divided into normal group, asthma group and IL-10 group, with 10 rats in each group. Asthma group and IL-10 group were sensitized by intraperitoneal injection of 1g.L-1 ovalbumin (VOA), and a mouse asthma model was induced by atomization inhalation with 10g.L-1VOA20mL. Normal group without any treatment. Asthma group before intranasal instillation of 9g.L-1 saline before each challenge, IL-10 group before each challenge with IL-10 (0.2μg.L-1) of 9g.L-1 saline 0.1mL Instillation of the nose. After the last challenge, the serum was separated from the eyeball of mice and the level of serum IgE was measured by ELISA. Take the lung tissue biopsy. Results The pathological changes in the asthmatic mice were significantly different from those in other groups. Inflammatory cells (mainly eosinophils and neutrophils) infiltrating into the bronchi and perivascular cells were obvious. The mucosal and submucosal edema were obvious. Mucus and mucus plug. The IL-10 group and the normal group no significant difference: a clear alveolar structure, alveolar and bronchial lumen without exudate, compared with the asthma group were significantly different. The levels of IgE in the normal group, asthma group and IL-10 group were (5.219 ± 0.427) μg.L-1, (6.633 ± 0.145) μg.L-1 and (5.503 ± 0.566) μg.L- The level of serum IgE in asthma group was significantly lower than that in normal group and IL-10 group (P <0.05). There was no significant difference between IL-10 group and normal group (P> 0.05). Conclusion IL-10 local drip therapy can inhibit lung allergic inflammation in asthmatic mice, improve respiratory symptoms and reduce serum IgE levels in asthmatic mice.
