目的探讨2型糖尿病患者外周血单核细胞中嘌呤能受体(P2X7R)的表达变化及其与血清C-反应蛋白(CRP)和细胞因子(TNF-α、IL-10、IL-1β)变化的相关性。方法选取38例2型糖尿病患者作为实验组(其中20例CRP正常患者,18例CRP升高患者),20例健康体检者为正常对照组,通过免疫比浊法检测CRP,酶联免疫吸附法(ELISA)检测血清细胞因子(TNF-α、IL-10、IL-1β)水平;并采用实时荧光定量PCR(RT-PCR)法检测外周血单核细胞P2X7R的mRNA表达水平(以β-actin为内参)。结果 2型糖尿病组患者的CRP、TNF-α及IL-1β水平显著高于正常对照组,P2X7R基因的表达水平也显著高于正常对照组,差异均有统计学意义(P<0.05)。CRP升高2型糖尿病患者的TNF-α、IL-1β的水平明显高于CRP正常2型糖尿病组患者,而CRP升高2型糖尿病组患者的IL-10水平显著低于其他2组,差异均有统计学意义(P<0.05)。结论单核细胞表达的P2X7R基因是2型糖尿病患者的重要危险因子,与CRP水平无关,在2型糖尿病的发生及进展过程中可能具有重要作用。
Objective To investigate the expression of P2X7R in peripheral blood mononuclear cells of patients with type 2 diabetes mellitus (T2DM) and its relationship with the changes of serum C-reactive protein (CRP) and cytokines (TNF-α, IL-10 and IL- Relevance. Methods Thirty-eight patients with type 2 diabetes mellitus (20 normal subjects and 18 patients with elevated CRP) were selected as experimental group and 20 healthy subjects as normal control group. Results The levels of CRP, TNF-α and IL-1β in patients with type 2 diabetes mellitus were significantly higher than those in normal control group, and the expression level of P2X7R gene was also significantly higher in patients with type 2 diabetes mellitus than in controls (P <0.05). The levels of TNF-α and IL-1β in patients with type 2 diabetes mellitus with CRP increase were significantly higher than those with type 2 diabetes mellitus with CRP, while those with type 2 diabetes mellitus were significantly lower than those in patients with type 2 diabetes mellitus All were statistically significant (P <0.05). Conclusion The P2X7R gene expressed in monocytes is an important risk factor for type 2 diabetes, which has nothing to do with the level of CRP. It may play an important role in the development of type 2 diabetes mellitus.
