将手术切除的新鲜脑胶质瘤组织制备成单细胞悬液并进行短期体外培养,采取用流式细胞术测定瘤细胞的DNA指数(DI)、细胞周期分布及细胞分裂增殖指数(PI)等指标。结果发现,(1)脑胶质瘤Ⅲ、Ⅳ级之间的DNA异倍体率差异无显著性,但与其它各组差异有显著性(P<0.05)。(2)对照组的DI值与脑胶质瘤Ⅰ级差异无显著性,但与其它各组差异有高度显著性(P<0.01)。(3)对照组的S期与脑胶质瘤Ⅰ、Ⅱ、级之间差异无显著性,但与Ⅲ、Ⅳ级差异有高度显著性(P<0.01)。(4)对照组的PI值与肿瘤各组差异均有高度显著性(P<0.01)。提示脑胶质瘤细胞核DNA的流式细胞术分析能较准确的判定肿瘤的恶性程度,对脑胶质瘤患者的诊断、术后辅助治疗及预后判断均有重要意义。
The freshly-resected glioma tissue was prepared into single-cell suspension and cultured in vitro for short periods. The DNA index (DI), cell cycle distribution and proliferation index (PI) of tumor cells were determined by flow cytometry index. The results showed that (1) There was no significant difference in DNA aneuploidy between grade III and grade IV glioma, but there was significant difference with other groups (P <0.05). (2) There was no significant difference in DI value between the control group and glioma grade Ⅰ, but it was highly significant with other groups (P <0.01). (3) There was no significant difference between S phase and gliomas grade Ⅰ, Ⅱ, and grade in control group, but significant difference between grade S and grade Ⅳ (P <0.01). (4) The PI value of the control group was significantly different from that of the tumor groups (P <0.01). It is suggested that the flow cytometry analysis of nuclear DNA in glioma cells can determine the degree of malignancy of the tumor more accurately and is of great significance for the diagnosis of glioma patients, postoperative adjuvant therapy and prognosis.
