为了研究四氯化碳(CCL4)联合二乙基亚硝胺(DEN)对小鼠(Mus musculus domesticus)精巢的毒性作用。将昆明小鼠随机分为对照组与实验组,各12只个体。实验组每周2次腹腔注射含有15%CCl4的花生油溶液,同时自由饮用0.07‰的DEN溶液,连续诱导8周。对照组每周2次注射花生油,饮用蒸馏水。分别于第3、6、8周处死实验组及对照组小鼠各4只,对其精巢组织进行常规固定、包埋、切片、H.E染色,观察精巢的形态及精子数量的变化。与实验组比较,对照组小鼠体重明显增加,且实验组与对照组差异显著;与对照组相比,实验组睾丸系数以及精子存活率均显著减少。对照组小鼠精巢都具有正常的生精小管结构,管腔中存在大量精子;第3周,实验组小鼠的精巢与对照组类似,生精小管较为完整,生精细胞稍有散乱;第6周,实验组生精小管变形,生精细胞排列疏松,精子数目减少且畸形;第8周,实验组生精小管残缺不全,生精细胞和精子散乱排列,精子数量大大减少且变形更为严重。实验说明,CCl4联合DEN能够造成小鼠精巢组织的损害以及精子数目的减少和畸变。
In order to investigate the toxic effects of carbon tetrachloride (CCL4) combined with diethylnitrosamine (DEN) on testis in Mus musculus domesticus. Kunming mice were randomly divided into control group and experimental group, 12 individuals each. Experimental group 2 times per week by intraperitoneal injection of 15% CCl4 containing peanut oil solution, while free drinking 0.07 ‰ of DEN solution, continuous induction of 8 weeks. The control group was injected twice a week peanut oil, drinking distilled water. At the 3rd, 6th and 8th week respectively, 4 mice in each experimental group and control group were sacrificed. The testis tissues were routinely fixed, embedded, sectioned and stained with H.E. The morphology of the testis and sperm count were observed. Compared with the experimental group, the body weight of the control group increased significantly, and the difference between the experimental group and the control group was significant. Compared with the control group, the testicular coefficient and sperm survival rate of the experimental group were significantly decreased. The testis in the control group had normal seminiferous tubule structure and a large number of sperm in the lumen. At the third week, the testis in the experimental group was similar to the control group, the seminiferous tubules were relatively complete and the spermatogenic cells were slightly scattered. After 6 weeks, the seminiferous tubules were deformed, the spermatogenic cells were loosely arranged and the number of spermatozoa was decreased and deformed. In the 8th week, the seminiferous tubules were incomplete, the spermatogenic cells and spermatozoon were arranged randomly, the sperm quantity was greatly reduced and the deformation was more serious. Experiments show that CCl4 combined with DEN can cause damage to the mouse testis tissue and sperm count reduction and distortion.
