目的比较异长春花碱(VRB)长循环脂质体、普通脂质体、重酒石酸盐注射液不同剂型的抗肿瘤作用。方法建立C57BL/6J小鼠Lewis肺癌模型,以肿瘤体积、小鼠生存时间、抑瘤率、组织病理切片为指标考察VRB不同制剂的抗肿瘤作用。结果与模型组(平均肿瘤体积为4 980 mm3)相比,VRB长循环脂质体组、普通脂质体组和重酒石酸盐注射液组小鼠造模30 d后的平均肿瘤体积分别为1 622、2 136、3 652 mm3(P<0.05、0.01),抑瘤率分别为66.29%、49.44%、29.21%(P<0.01);VRB长循环脂质体组小鼠生存时间明显延长;肿瘤组织病理切片观察可见各制剂组肿瘤细胞均出现不同程度的坏死,其中VRB长循环脂质体组出现肿瘤细胞核凝固坏死、核破碎溶解现象。结论在相同给药剂量下,VRB长循环脂质体的抗肿瘤作用明显优于其重酒石酸盐注射液。
Objective To compare the antitumor effects of different formulations of vincristine (VRB) long circulating liposomes, common liposomes and tartrate injection. Methods Lewis lung carcinoma model was established in C57BL / 6J mice. The anti-tumor effects of different preparations of VRB were investigated by tumor volume, survival time, tumor inhibition rate and histopathological sections. Results Compared with the model group (mean tumor volume of 4 980 mm3), the average tumor volume of mice in the VRB long-circulating liposome group, the normal liposome group and the tartrate injection group after 30 days of modeling was 1 622,2136 and 652 mm3 respectively (P <0.05, 0.01). The tumor inhibition rates were 66.29%, 49.44% and 29.21% respectively (P <0.01). The survival time of mice in VRB long- Histopathological examination showed that all the tumor cells in the preparation group showed different degrees of necrosis. Among them, the long-circulating VRB group showed tumor cell nuclear coagulation necrosis and nuclear fragmentation. Conclusions The antitumor effect of VRB long circulating liposomes is obviously better than that of the tartrate injection at the same dosage.
