目的:研究唑来膦酸联合Ghrelin对人成骨细胞hFOB1.19的作用。方法:以不同浓度唑来膦酸和Ghrelin处理成骨细胞,以MTT法检测其不同浓度和处理时间下细胞增殖情况,研究其最佳作用浓度;并以最佳作用浓度,单独和联合处理成骨细胞,检测成骨细胞在不同时间内碱性磷酸酶活性和骨钙素的表达量,研究成骨细胞的成骨活性。结果:唑来膦酸和Ghrelin对成骨细胞增殖均具有促进作用,其最佳作用浓度分别为10~(-10)mol/L和10~(-11)mol/L;该浓度下两者均可提高碱性磷酸酶活性和骨钙素表达量,但两者联合应用时碱性磷酸酶活性无明显差异,骨钙素表达量在作用72 h时有明显差异。结论:唑来膦酸和Ghrelin可以促进成骨细胞的增殖,提高碱性磷酸酶活性和骨钙素表达量,两者联合应用可能促进成骨细胞活性。
AIM: To investigate the effect of zoledronic acid combined with Ghrelin on human osteoblast hFOB1.19. Methods: The osteoblasts were treated with different concentrations of zoledronic acid and Ghrelin. The proliferation of osteoblasts was detected by MTT assay at different concentrations and treatment time. The optimal concentrations of zoledronic acid and Ghrelin were studied. Osteoblast, osteoblasts were detected at different times of alkaline phosphatase activity and osteocalcin expression, osteoblast osteogenic activity. Results: Both zoledronic acid and Ghrelin promoted the proliferation of osteoblasts, and the optimal concentrations of zoledronic acid and Ghrelin were 10 ~ (-10) mol / L and 10 ~ (-11) mol / L, respectively. Can increase alkaline phosphatase activity and osteocalcin expression, but the combination of the two had no significant difference in alkaline phosphatase activity, osteocalcin expression in the role of 72 h were significantly different. Conclusion: Zoledronic acid and Ghrelin can promote the proliferation of osteoblasts and increase the activity of alkaline phosphatase and the expression of osteocalcin, and the combination of them may promote the activity of osteoblasts.
