目的用化学指标结合药效学指标优选丹参饮的提取工艺。方法分别采用水提、超临界CO_2提取、超临界CO_2提取结合药渣水提、超临界CO_2提取结合药渣乙醇提取制备丹参饮提取物,以指标化学成分提取率为考察对象,采用均匀设计安排实验,优选各提取工艺参数,并观察丹参饮各提取物对急性心肌缺血小鼠心电图(ECG)和血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)活性的影响。结果丹参饮各提取物对异丙肾上腺素(ISO)诱导的小鼠缺血性ECG改变有明显的拮抗作用,能显著降低心肌缺血损伤时血清CK、LDH活性,对缺血心肌具有良好的保护作用,且以超临界CO_2提取结合药渣乙醇提取制得的提取物的药效最佳。结论超临界CO_2提取结合药渣乙醇提取为最佳提取工艺,所得丹参饮提取物的药效作用最佳。
Objective To optimize the extraction process of Salvia miltiorrhiza with chemical indicators and pharmacodynamics indexes. Methods The aqueous extract, supercritical CO_2 extraction, supercritical CO_2 extraction combined with dregs water extraction, supercritical CO_2 extraction combined with dregs ethanol extraction preparation of Salvia extract, index chemical composition extraction rate as the study object, using uniform design arrangements Experiments were conducted to optimize the extraction process parameters and to observe the effect of each extract of Danshen on ECG and serum creatine kinase (CK) and lactate dehydrogenase (LDH) activity in acute myocardial ischemia mice. Results The extracts from Danshen Yin could obviously antagonize the ischemic ECG changes induced by isoproterenol (ISO) in mice, and could significantly reduce the activity of serum CK and LDH in myocardial ischemic injury and had good Protective effect, and the supercritical CO2 extraction combined with dregs extracted ethanol extract obtained the best efficacy. Conclusion The supercritical CO_2 extraction combined with dregs ethanol extraction is the best extraction process, and the obtained extract of Radix Salviae Miltiorrhizae is the best.
