目的:建立葛根清热颗粒中3种活性成分的含量测定方法。方法:采用高效液相色谱法,甲醇A-0.01mol.L-1磷酸二氢钾溶液B(磷酸调pH2.5);流速1.0mL.min-1;检测波长210nm。梯度洗脱程序A:0~5min,10%;5~30min,10%~30%;30~40min,30%。结果:3种化学成分均达到较好地分离,峰形良好,葛根素、盐酸麻黄碱、芍药苷的线性范围分别为0.822~1.918,0.468~1.092,0.061~0.143μg;平均加样回收率分别为99.8%,99.7%,99.9%;RSD分别为1.3%,1.3%,2.0%。结论:该方法操作简单,准确,重复性好,可用于葛根清热颗粒的质量标准。
Objective: To establish a method for the determination of three active ingredients in Pueraria radix granules. Method: High performance liquid chromatography, methanol A-0.01mol.L-1 potassium dihydrogen phosphate solution B (phosphoric acid pH 2.5); flow rate 1.0mL.min-1; detection wavelength 210nm. Gradient elution program A: 0~5min, 10%; 5~30min, 10%~30%; 30~40min, 30%. Results: The three chemical components all achieved good separation and good peak shape. The linear ranges of puerarin, ephedrine hydrochloride and paeoniflorin were 0.822 to 1.918, 0.468 to 1.092, and 0.061 to 0.143 μg, respectively; It was 99.8%, 99.7%, and 99.9%; RSD was 1.3%, 1.3%, and 2.0%, respectively. Conclusion: This method is simple, accurate and reproducible. It can be used for the quality standard of Pueraria radix granules.
