[目的]探讨多维分离策略从半枝莲中分离野黄芩苷的可行性。[方法]采用具有互补分离机理的大孔树脂法及有机溶剂萃取法,以及高速逆流色谱法在优化的条件下分离半枝莲中的野黄芩苷类成分,通过高效液相色谱分析检测,质谱法定性。[结果]采用D101大孔树脂粗分离时,20%乙醇水洗脱组分固形物含量较高。采用有机溶剂萃取法溶剂系统为正丁醇∶乙酸乙酯∶水(1∶1∶2,v/v/v)时,水相中主成分含量较高。高速逆流色谱法溶剂系统为正丁醇∶水(1∶1,v/v)时,能达到较好的分离效果,对主成分峰进行鉴定后确定为野黄芩苷。[结论]结合多维分离策略,能够从半枝莲中高效分离制备得到野黄芩苷。
[Objective] The research aimed to discuss the feasibility of separation of wild baicalin from Scutellaria barbata with multidimensional separation strategy. [Method] With the separation mechanism of macroporous resin and organic solvent extraction, as well as high-speed countercurrent chromatography in the conditions of Scutellaria barbata separation of components, by high performance liquid chromatography analysis, mass spectrometry Legal nature. [Result] The content of solid fraction eluted by 20% ethanol water was higher when crude resin was separated by D101 macroporous resin. The organic solvent extraction solvent system is n-butanol: ethyl acetate: water (1: 1: 2, v / v / v), the main component of the water content higher. High-speed countercurrent chromatography with n-butanol: water (1: 1, v / v) as solvent system can achieve a better separation effect, and the main component peaks were identified as wild baicalin. [Conclusion] Combined with multidimensional separation strategy, Scutellaria baicalensis Georgi could be isolated and prepared efficiently from Scutellaria barbata.
