目的;为探讨局部脑缺血/再灌流中肿瘤坏死因子-α(TNF-α)的动态变化及其与白细胞浸润的关系。方法:采用大鼠大脑中动脉梗死模型,分别用免疫组织化学方法检测TNF-α、酶组化方法检测髓过氧化物酶(MPO)的动态变化,用图像分析进行定量分析。结果:TNF-α在缺血组(I)和缺血再灌流组(IR)0.5h表达增多,12h达高峰,与对照组比较差异非常显著(P<0.01).持续至120h;1组12h、IR组0.5hMPO明显增多,高峰在24h(P<0.01);I组TNF-α与APO有非常显著相关性(P<0.01),IR组TNF-α与MPO有显著相关性(P<0.05),证明局部脑缺血/再灌流损伤中TNF-α表达增多.其动态变化与损伤时间和局部白细胞浸润关系密切,结论:TNF-α是缺血性脑损伤的重要因素。
Objective To investigate the dynamic changes of tumor necrosis factor-α (TNF-α) and its relationship with leukocyte infiltration in focal cerebral ischemia / reperfusion. Methods: The model of middle cerebral artery occlusion in rats was established. The expression of TNF-α was detected by immunohistochemical method. The changes of myeloperoxidase (MPO) were detected by enzymatic histochemical method. Quantitative analysis was performed by image analysis. Results: The expression of TNF-α increased at 0.5 hour in ischemic group and IR group, reaching the peak at 12 hours, which was significantly different from the control group (P <0.01). (P <0.01). The level of TNF-α in the IR group was significantly higher than that in the APO group (P <0.01) And MPO (P <0.05), demonstrating the increased expression of TNF-α in focal cerebral ischemia / reperfusion injury. The dynamic changes were closely related to the injury time and the local leukocyte infiltration. Conclusion: TNF-α is an important factor of ischemic brain injury.
