目的探讨胃泌素和胃泌素受体拮抗剂丙谷胺对胃癌细胞株SGC-7901增殖和环氧化酶2(COX-2)表达的调控作用。方法SGC-7901细胞接种于含10%胎牛血清的RPMI-1640培养液中常规培养24h后,换无血清培养基继续培养24h,加入胃泌素(终浓度为0.1、1、10、100nmol/L)或胃泌素(100nmol/L)加丙谷胺(终浓度为1、10、100、1000μmol/L),分别培养24、48和72h;MTT比色分析细胞增殖,Western blot检测COX-2蛋白表达。结果胃泌素呈时间和剂量依赖性地促进SGC-7901细胞增殖,而丙谷胺抑制了胃泌素诱导的细胞增殖。胃泌素增加COX-2在SGC-7901细胞的表达,丙谷胺有效抑制胃泌素诱导的COX-2表达。结论胃泌素通过胃泌素受体诱导COX-2表达促进胃癌细胞增殖。
Objective To investigate the regulation of gastrin and gastrin receptor antagonist proglumide on the proliferation and cyclooxygenase 2 (COX-2) expression in gastric cancer cell line SGC-7901. Methods SGC-7901 cells were inoculated into RPMI-1640 culture medium containing 10% fetal bovine serum for 24 hours and then cultured in serum-free medium for 24 hours. Gastrin (final concentration 0.1,1,10,100 nmol / L) or gastrin (100nmol / L) plus proglumide (final concentration of 1, 10, 100 and 1000μmol / L) for 24, 48 and 72h respectively. MTT colorimetric assay was used to detect cell proliferation. Western blot was used to detect COX- 2 protein expression. Results Gastrin promoted the proliferation of SGC-7901 cells in a time- and dose-dependent manner, whereas proglumide inhibited gastrin-induced cell proliferation. Gastrin increased COX-2 expression in SGC-7901 cells, and proglumide effectively inhibited gastrin-induced COX-2 expression. Conclusion Gastrin promotes COX-2 expression through gastrin receptor and promote the proliferation of gastric cancer cells.
