目的 :构建乙型肝炎病毒 (HBV ) adr亚型基因疫苗 p CMV- S2 .S(PS) ,观察其诱导小鼠所产生的特异性体液和细胞免疫应答。方法 :将 PS注射于 C5 7BL / 6小鼠胫骨前肌内 ,应用 EL ISA方法检测不同时间小鼠血清中抗 HBs抗体以及采用 51Cr释放实验检测淋巴细胞杀伤功能。结果 :注射基因疫苗 1周后 ,血清中抗 HBs抗体转阳 ,4周后抗体达到高峰 ,并以高水平维持至少 8周 ;第 12周时 PS诱导小鼠产生了良好的 HBV特异的细胞免疫应答 (P<0 .0 5 )。结论 :所构建的 HBV adr亚型基因疫苗 PS能够诱导小鼠产生较好的体液和细胞免疫应答
Objective: To construct the hepatitis B virus (HBV) adr subtype gene vaccine, CMV-S2.S (PS), and to observe the specific humoral and cellular immune responses induced by it in mice. Methods: PS was injected into the tibialis anterior muscle of C5 7BL / 6 mice. Anti-HBs antibodies in sera of mice were detected by ELISA at different time points. The cytotoxicity of lymphocytes was detected by 51Cr release assay. Results: Anti-HBs antibody in the serum turned positive after 1 week of injection of the gene vaccine. After 4 weeks, the antibody peaked and maintained at a high level for at least 8 weeks. PS-induced mice produced good HBV-specific cellular immunity Response (P <0. 05). Conclusion: The constructed HBV adr subtype gene vaccine PS can induce mice to produce better humoral and cellular immune responses
