目的:探讨内皮素-1与血管平滑肌细胞增殖的关系。方法:用噻唑蓝比色法观察内皮素-1对血管平滑肌细胞增殖的影响,流式细胞仪法观察对血管平滑肌细胞增殖周期的影响。结果:与对照组比较,内皮素-1明显促进血管平滑肌细胞增殖;促进平滑肌细胞DNA合成;促进平滑肌细胞从G0期向S期的转变,G0/G1期细胞百分比明显降低,而S期细胞百分比增加。结论:内皮素-1对血管平滑肌细胞增殖有明显的促进作用。
Objective: To investigate the relationship between endothelin-1 and proliferation of vascular smooth muscle cells. Methods: The effect of endothelin-1 on the proliferation of vascular smooth muscle cells was observed by thiazolyl blue colorimetric assay. The effect of endothelin-1 on the proliferation of vascular smooth muscle cells was observed by flow cytometry. Results: Compared with the control group, endothelin-1 significantly promoted the proliferation of vascular smooth muscle cells; promoted the DNA synthesis of smooth muscle cells; promoted the transition from G0 phase to S phase of smooth muscle cells, while the percentage of cells in G0 / G1 phase decreased significantly; while the percentage of S phase cells increase. Conclusion: Endothelin-1 can significantly promote the proliferation of vascular smooth muscle cells.
