当用血吸虫童虫进行免疫荧光试验时,由于童虫较大,在不作切片的情况下,会出现某些困难,主要是童虫固定较难。作者在固定过程中,因采用了明胶液加重铬酸钾的方法而获得了满意的结果。方法如下:将5%重铬酸钾溶液1ml加到200ml的0.25%(W/V)明胶溶液中,置水浴加热5~10分钟,直至完全混匀。待冷至室温,然后一滴滴地加于玻片上,于室温干燥。将收集的血吸虫童虫用PBS(0.15M,pH7.4)洗涤2次后,以1000转/分速度离心1分钟,弃去上清液。在小试管内将童虫再混悬于2ml PBS
When using immunofluorescence with schistosomiasis, there are some difficulties that can arise in the absence of slicing due to the larger size of the squirrels, mainly because it is harder to fix the squirrels.
