目的 :探讨各种血栓形成相关性疾病患者的P -选择素的表达变化及其临床意义。方法 :用异硫氰酸荧光素 (FITC)标记抗P -选择素单抗SZ - 5 1-IgG和流式细胞术 (FCM)分析了 42例糖尿病患者、33例高血脂患者、2 3例脑梗塞患者和 2 0例正常人的血小板膜P -选择素表达 ,并与间接FCM和酶标记免疫吸附分析法(ELISA)作比较。结果 :FCM测得的血小板膜上P -选择素表达阳性率 ,在糖尿病组为 2 3 92 %± 15 83% ,高血脂组为 18 34 %± 9 46 % ,脑梗塞组为 19 32 %± 10 38% ,均显著高于正常对照 3 38%± 1 11% (P值均小于 0 0 1)。间接FCM测定血小板膜P -选择素表达在糖尿病和脑梗塞患者中均显著增高 ,ELISA法测得的血浆中P -选择素在糖尿病、高血脂和脑梗塞患者也显著高于正常对照。结论 :FITC标记的SZ - 5 1-IgG可用于FCM ,可成为测定血小板活化的一个新的敏感的方法
Objective: To investigate the changes of P-selectin in patients with various thrombosis-related diseases and its clinical significance. Methods: Forty - two diabetic patients, 33 hyperlipidemia patients and 23 patients with hyperlipidemia were analyzed by fluorescein isothiocyanate (FITC) labeled anti - P - selectin monoclonal antibody SZ - 5 1 - IgG and flow cytometry (FCM) The platelet membrane P - selectin expression in 20 patients with cerebral infarction and 20 normal controls was compared with indirect FCM and enzyme - linked immunosorbent assay (ELISA). Results: The positive rate of P - selectin in platelet membrane measured by FCM was 2292% ± 1583% in diabetic group, 18 34% ± 9 46% in hyperlipidemia group and 19 32% ± in cerebral infarction group 10 38%, both significantly higher than the control 38% ± 1111% (P values were less than 0 0 1). Indirect FCM measured platelet membrane P - selectin expression in patients with diabetes mellitus and cerebral infarction were significantly increased, ELISA measured plasma P - selectin in patients with diabetes, hyperlipidemia and cerebral infarction was significantly higher than the normal control. CONCLUSION: FITC-labeled SZ-5 1-IgG can be used in FCM as a new and sensitive method of platelet activation
