目的观察实验大鼠逐渐形成的前列腺炎和与之伴随的尿动力学改变。方法成年Wistar雄性大鼠50只,随机分为5组,每组10只,5组分别为假手术组、E_2 0.25mg/kg 15d组、E_2 0.25mg/kg 30d组、E_2 0.125mg/kg组和模型对照组。除假手术组外,其他4组大鼠均在无菌条件下去势,术后第2d分别连续给予稀释于花生油的苯甲酸雌二醇(E_2 0.25mg/kg·d~(-1))15d及30d和苯甲酸雌二醇(E_2 0.125mg/kg·d~(-1))30d,模型对照组注射等量的花生油30d,假手术组仅皮下注射等量的生理盐水30d。于末次给药后第2天在麻醉下行尿动力学检查,最后作实验大鼠前列腺的组织病理学分析。结果雌激素可诱导大鼠发生慢性非细菌性前列腺炎,E_2 0.25mg/kg给药30d组排尿阈值压增加,顺应性及膀胱容量降低,与对照组及假手术组相比,差异有显著性(P<0.05), E_2 0.25mg/kg 15d组和E_2 0.125mg/kg 30d组与对照组及假手术组相比尿动力学指标无显著性差异(P>0.05)。结论慢性非细菌性前列腺炎动物模型存在与腺体炎症相关的尿动力学改变。
Objective To observe the progressive formation of prostatitis and the accompanying urodynamic changes in experimental rats. Methods Fifty adult Wistar male rats were randomly divided into 5 groups with 10 rats in each group. The rats in each group were sham operation group, E_2 0.25 mg / kg 15 d group, E 2 0.25 mg / kg 30 d group, E 2 0.125 mg / kg group And model control group. Except for the sham group, the other four groups of rats were castrated under aseptic conditions. Serum estradiol benzoate (E 2 0.25 mg / kg · d -1) diluted in peanut oil was continuously administered on the 2nd day after operation for 15 days (E 0.125mg / kg · d -1) for 30 days, and the model control group received the same amount of peanut oil for 30 days. The rats in the sham operation group were injected subcutaneously with normal saline for 30 days. On the second day after the last administration, urodynamic tests were performed under anesthesia, and finally the histopathological analysis of the experimental rats was performed. Results Estrogen induced chronic nonbacterial prostatitis in rats. E_2 0.25 mg / kg administered 30 d increased urinary threshold pressure, decreased compliance and bladder capacity, compared with the control group and the sham operation group, the difference was significant (P <0.05). There was no significant difference in urodynamic parameters between E_2 0.25mg / kg 15d group and E_2 0.125mg / kg 30d group and control group and sham operation group (P> 0.05). Conclusion The animal models of chronic nonbacterial prostatitis have urodynamic changes associated with glandular inflammation.
