1991年Kromer报道了一种用拮抗剂而不测pA_2值进行受体分型的新方法,有意避免测定拮抗剂对所研究组织的亲和力。该方法适用于电场刺激引起的内源性激动剂的释放实验。本文提出一个更接近于经典测定pA_2值的方法。当用激动剂单一浓度产生效应时,能获得一条相应的拮抗剂浓度-效应曲线,诚然该拮抗剂的绝对pA_2值不能由此结果求得,但能计算其相对pA_2值。假若激动剂A与拮抗剂Ⅰ竞争同一受体,那么,被激动剂占领的受体部分是:
Kromer (1991) reported a new method of receptor typing using antagonist rather than pA2 values, deliberately avoiding the need to determine the affinity of the antagonist for the tissue being studied. The method is suitable for the release of endogenous agonists caused by electric field stimulation. This paper presents a method that is closer to the classical determination of pA_2 values. When a single concentration of agonist produces an effect, a corresponding antagonist concentration-response curve can be obtained. It is true that the absolute pA_2 value of the antagonist can not be calculated from this result but the relative pA_2 value can be calculated.
