目的研究西妥昔单抗对乳腺肿瘤细胞生长的影响。方法 MDA-MB-231细胞在2μg/mL西妥昔单抗刺激12h和24h后,收集细胞进行裂解。等量蛋白进行蛋白印迹法检测integrinβ5表达。软琼脂克隆形成实验检测西妥昔单抗对细胞生长的影响。结果西妥昔单抗明显降低integrinβ5蛋白表达。MDA-MB-231细胞在西妥昔单抗的作用下降低了克隆形成数量。结论西妥昔单抗能够下调integrinβ5表达,抑制乳腺癌细胞锚定非依赖生长,提示西妥昔单抗可能是重要的乳腺癌治疗靶向药物。
Objective To study the effects of cetuximab on the growth of breast tumor cells. Methods After MDA-MB-231 cells were stimulated with 2μg / mL cetuximab for 12h and 24h, cells were harvested and lysed. The same amount of protein was detected by western blotting for the expression of integrinβ5. Soft agar colony formation assay cetuximab on cell growth. Results Cetuximab significantly decreased integrinβ5 protein expression. MDA-MB-231 cells reduced the number of clones formed by cetuximab. Conclusion Cetuximab can down-regulate the expression of integrinβ5 and inhibit the anchorage-independent growth of breast cancer cells, suggesting that cetuximab may be an important therapeutic target for breast cancer.
