目的探讨补阳还五汤对大脑中动脉栓塞后不同时间大鼠脑内微小核糖核酸(miRNA)表达的影响。方法建立大脑中动脉栓塞的脑缺血模型,芯片方法观察miRNA的表达差异,RT-PCR进行验证。结果与3天模型组相比,补阳还五汤组miRNA有2个上调,1个下调;与15天模型组相比,补阳还五汤组miRNA有5个上调,4个下调。随后,选取3个miRNA使用RT-PCR验证各组大鼠脑内miRNA差异表达,同芯片结果相符。结论补阳还五汤对脑缺血后3天和15天miRNA均能产生影响,提示补阳还五汤对脑缺血急性期和修复期都具有治疗作用。另外,相比较而言,补阳还五汤对脑缺血后15天miRNA影响的数量更多,可能其促进修复的作用更具有优势。
Objective To investigate the effect of Buyang Huanwu Decoction on the expression of miRNAs in rat brain at different time after middle cerebral artery occlusion. Methods The model of cerebral ischemia induced by middle cerebral artery occlusion was established. The difference of miRNA expression was observed by microarray and verified by RT-PCR. Results Compared with the 3-day model group, miRNAs of Buyang Huanwu Decoction group were up-regulated and down-regulated. Compared with the 15-day model group, miRNAs of Buyang Huanwu Decoction group were increased by 5 and down-regulated by 4. Subsequently, three miRNAs were selected and used to verify the differential expression of miRNAs in the brain of each group by RT-PCR, which was in good agreement with the results of the microarray. Conclusion Buyang Huanwu decoction can affect the miRNAs on the 3rd and 15th days after cerebral ischemia, suggesting that Buyang Huanwu decoction has a therapeutic effect on the acute and repaired phases of cerebral ischemia. In addition, in comparison, the effect of Buyang Huanwu Decoction on the number of miRNAs 15 days after cerebral ischemia more likely to promote the role of repair more advantages.
