LycB(番茄红素β-环化酶 )基因是类胡萝卜素生物合成过程中关键酶之一,位于合成代谢的重要分支点上,直接影响β-胡萝卜素的合成。以番茄子叶、茎段作为转化受体,通过农杆菌介导法,利用LycB基因转化重要果菜两用作物番茄,对影响转化的几个因素进行了研究。结果表明,转化受体经预培养比未预培养有利于农杆菌的侵染转化;侵染时间子叶以 10min、茎段以 15min为宜;农杆菌菌液浓度OD600以 0 7为佳;添加AS可提高抗性愈伤的诱导率,AS的添加以 100μmol/L效果较好;延迟筛选有利于提高抗性愈伤诱导率。
LycB (lycopene β-cyclase) gene is one of the key enzymes in the process of carotenoid biosynthesis. It is located at an important branch of anabolism and directly affects the synthesis of β-carotene. Using tomato cotyledons and stem segments as transformation receptors, Agrobacterium-mediated transformation of LycB gene was used to transform fruit and vegetable dual-purpose crops and tomatoes. Several factors affecting transformation were studied. The results showed that the transformants were preincubated better than the non-preadulticted ones for the transformation of Agrobacterium. The cotyledons were infested at 10 min and 15 min respectively, and the OD600 of Agrobacterium was better than 0 7. Can increase the induction rate of resistant callus, the effect of adding AS at 100 μmol / L is better; and the delayed screening is beneficial to improving the resistance callus induction rate.
