目的:制定痔疮熏洗液的质量标准。方法:用薄层色谱法定性鉴别功劳木、苦参和黄柏;采用高效液相色谱法同时测定痔疮熏洗液中盐酸小檗碱和苦参碱含量,色谱柱:WondaSil Superb C18(4.6 mm×250 mm,5μm),流动相:乙腈-甲醇-磷酸盐缓冲液(pH6.8)-三乙胺(22∶22∶70∶0.1),检测波长:220 nm,流速:1.0ml·min-1,柱温:30℃。结果:薄层色谱斑点清晰,分离度好,阴性无干扰;盐酸小檗碱和苦参碱的线性范围分别为5.8~92.8 mg·L-1(r=0.999 9),6.4~102.4 mg·L-1(r=0.999 9),平均加样回收率(n=9)分别为98.17%,97.57%,RSD分别为1.57%,1.42%。结论:定性定量分析方法快速、准确、专属性强,本法可控制痔疮熏洗液的质量。
Objective: To develop hemorrhoids smoked liquid quality standards. Methods: The qualitative identification of Koumu, Sophora flavescens and Phellodendron amurense by TLC; the content of berberine hydrochloride and matrine in Hemorrhoid Washing Lotion were determined simultaneously by high performance liquid chromatography. Column: WondaSil Superb C18 (4.6 mm × 250 mm, 5μm). The mobile phase was acetonitrile-methanol-phosphate buffer (pH6.8) -triethylamine (22:22:70:0.1). The detection wavelength was 220 nm and the flow rate was 1.0 ml · min -1 Column temperature: 30 ° C. Results: The spots of TLC were clear, the resolution was good and the negative was no interference. The linear range of berberine hydrochloride and matrine were 5.8 ~ 92.8 mg · L-1 (r = 0.999 9), 6.4 ~ 102.4 mg · L -1 (r = 0.999 9). The average recoveries (n = 9) were 98.17% and 97.57%, respectively, with RSDs of 1.57% and 1.42%, respectively. Conclusion: The method of qualitative and quantitative analysis is rapid, accurate and specific. The method can control the quality of hemorrhoids smoked liquid.
