目的:观察中药复方加味四逆散水煎剂对血吸虫病小鼠肝纤维化形成的影响。方法:NIH白鼠经皮肤人工感染日本血吸虫尾蚴造模,随机分为4组:B组(模型组);C组(吡喹酮组);D组(吡喹酮+加味四逆散组);E组(吡喹酮+秋水仙碱组),同时设A组(正常对照组)。病理证实小鼠肝组织纤维化形成后的第21 d开始给药治疗,第45 d时各治疗组分别予以吡喹酮300 mg·kg-1一次性灌胃进行杀虫处理,第78 d处死小鼠,取肝组织切片HE染色观察病理变化,免疫组化法检测肝组织Ⅰ型、Ⅲ型胶原。结果:各组小鼠肝组织Ⅰ型胶原积分光密度均值比较:加味四逆散组明显低于模型组、吡喹酮组及秋水仙碱组(P<0.05)。加味四逆散组、秋水仙碱组Ⅲ型胶原的表达明显低于模型组、吡喹酮组(P<0.05)。结论:中药加味四逆散联合吡喹酮能改善血吸虫病小鼠肝组织病理,降低小鼠肝组织Ⅰ型胶原、Ⅲ型胶原的表达。
Objective: To observe the effect of Jiawei Sini Decoction on hepatic fibrosis in mice with schistosomiasis. Methods: NIH mice were inoculated with cercariae of Schistosoma japonicum by artificial infection on the skin. They were randomly divided into 4 groups: group B (model group), group C (praziquantel group), group D (praziquantel + Jiawei Sini Powder group) Group E (praziquantel + colchicine group), while A group (normal control group). Pathology confirmed the formation of liver fibrosis in mice on the first 21 d after the start of treatment, the first 45 d of each group were treated with praziquantel 300 mg · kg-1 intragastric administration of insecticide treatment, died on the 78th Mice were sacrificed and liver sections were stained with HE for pathological changes. Immunohistochemistry was used to detect type Ⅰ and type Ⅲ collagen in liver tissues. Results: Compared with model group, praziquantel group and colchicine group (P <0.05), the average optical density of type I collagen in liver tissue of each group was significantly lower than that in the model group. The expression of type Ⅲ collagen in Jiawei Sini San group and colchicine group was significantly lower than that in model group and praziquantel group (P <0.05). CONCLUSION: Traditional Chinese medicine Sini Decoction combined with praziquantel can improve liver histopathology of mice with schistosomiasis and reduce the expression of collagen type Ⅰ and type Ⅲ collagen in mice liver.
