本文报道用离子对反相液相色谱—电化学检测法同时测定大鼠脑内单胺递质及其代谢产物。本法采用高氯酸沉淀组织蛋白后上清液直接进佯技术,能使NE、E、DA、5-HT及它们的主要代谢产物MHPG、DHPG、HVA、DOPAC、5-HIAA与内标异丙肾上腺奈等十种物质很好地同时分离。由此不仅能测定单胺类神经递质在脑内的含量,并能测定其转换率。方法简便快速,回收率较高且稳定。文中对色谱分离及检测的某些影响因素做了分析讨论,并测定了大鼠下丘脑内单胺递质及其代谢产物。
This paper reports the determination of monoamine neurotransmitters and their metabolites in rat brain by ion-pair reversed-phase liquid chromatography-electrochemical detection. This method using perchloric acid precipitation of tissue protein supernatant directly into the feces, can make NE, E, DA, 5-HT and their major metabolites MHPG, DHPG, HVA, DOPAC, 5-HIAA and internal standard Propranolol and other substances ten good separation at the same time. Thus not only can be measured monoamine neurotransmitters in the brain content, and can measure the conversion rate. The method is simple and fast, the recovery rate is high and stable. In this paper, some influencing factors of chromatographic separation and detection were analyzed and discussed, and monoamine neurotransmitters and their metabolites in rat hypothalamus were determined.
