本文用光敏生物素标记髓过氧化物酶(MPO)cDNA探针、以链亲和素胶体全为检测系统作细胞甩片原位杂交(ISH—SAG)检测了7株人白血病细胞系的MPO基因表达。结果发现,4株淋巴细胞系均不表达MPO基因,3株粒(单)细胞系均不同程度的表达MPO基因,HL—60细胞表达最强,KGI和U937表达较弱但却明显强于MPO组化染色。初步结果显示ISH—SAG检测MPO基因具有敏感、特异、简便的优点并能观察细胞形态,对于识别分化差的粒(单)细胞及白血病的分类诊断有重要意义。此外,作者还对ISH—SAG的部分实验条件作了讨论。
In this paper, myeloperoxidase (MPO) cDNA probe was labeled with photosensitizing biotin, and the MPO of seven human leukemia cell lines was detected by in situ hybridization (ISH-SAG) with streptavidin colloidal detection system. gene expression.
