目的探讨血浆中银的测定方法。方法血浆经体积分数为0.20%的曲拉通X-100溶液稀释后,在328.1 nm波长下,用石墨炉原子吸收光谱法直接测定血浆中银水平。结果本方法仪器检测限为0.14μg/L,定量限为0.48μg/L,最低检出浓度为1.40μg/L(取100.0μL血浆按10倍稀释计算);当银质量浓度在0.48~10.00μg/L时,线性关系良好,相关系数为0.999 4;加标回收率为97.00%~102.00%,批内精密度为1.42%~3.38%,连续3 d测定的批间精密度为1.84%~4.57%,血浆样品在4℃条件下可保存7 d。结论本方法线性关系良好、准确可靠、精密度高、操作简便、样品用量少,适用于大批量样品血浆中银的测定。
Objective To investigate the method of determination of silver in plasma. Methods Plasma was diluted with Triton X-100 solution (0.20% volume fraction) and plasma silver concentration was directly determined by graphite furnace atomic absorption spectrometry at a wavelength of 328.1 nm. Results The limit of detection was 0.14μg / L, the limit of quantification was 0.48μg / L and the minimum detectable concentration was 1.40μg / L (100.0μL of plasma was diluted by 10 times). When the silver concentration was between 0.48 and 10.00μg / L, the correlation coefficient was 0.999 4, the recoveries were 97.00% -102.00%, the intra-assay precision was 1.42% -3.88%, and the inter-assay precision was 1.84% -4.57 %, Plasma samples can be stored at 4 ℃ for 7 days. Conclusion The method has good linearity, accuracy and reliability, high precision, easy operation and less sample amount. It is suitable for the determination of silver in plasma of large sample.
