目的:观察益气活血剂中不同剂量黄芪组方对缺血性脑损伤大鼠的脑保护作用,以探讨黄芪适宜的使用剂量。方法:采用颈内动脉注射血栓的方法,复制多发梗死性脑缺血大鼠模型,观察益气活血剂中不同剂量黄芪(30g、60g、120g)组方对模型动物学习、记忆能力和脑组织胆碱酯酶(AchE)活性的影响。结果:60g黄芪配伍组模型大鼠学习、记忆能力明显改善,AchE含量降低,与空白对照组相仿,与模型对照组比较有极显著性意义。120g黄芪配伍组模型大鼠学习、记忆能力也得到改善,AchE含量降低,但其学习及24h记忆能力仍明显低于空白对照组。30g黄芪配伍组模型大鼠学习、记忆能力改善,AchE含量降低,但除72h记忆能力外,其余学习记忆能力均明显低于空白对照组,与模型对照组比较,24h记忆能力及AchE含量无显著性差别。结论:在治疗缺血性脑血管疾病的益气活血复方配伍中,黄芪的用量宜大,但随着其剂量的增大,反而减弱脑保护效应。其用量在人以60g/d左右为宜。
OBJECTIVE: To observe the cerebral protective effect of different doses of Astragalus membranaceus prescriptions in rats of Yiqi Huoxue Decoction on ischemic brain injury, so as to discuss the suitable dose of Radix Astragali. METHODS: The model of multiple infarction cerebral ischemia rat model was duplicated by injection of thrombus into the internal carotid artery. The effects of different doses of jaundice (30g, 60g, 120g) in the formula of learning Qi and promoting blood circulation on the learning, memory and brain tissues of model animals were observed. The effect of cholinesterase (AchE) activity. RESULTS: The learning and memory ability of 60g Huangqi compatibility group rats was significantly improved, and the AchE content was decreased. It was similar to the blank control group. Compared with the model control group, the results were extremely significant. The learning and memory ability of 120g Huangqi compatibility group rats also improved, AchE content decreased, but their learning and 24h memory ability were still significantly lower than the blank control group. The learning and memory ability of 30g Huangqi compatibility group rats were improved and the AchE content was decreased, but except for 72h memory capacity, the other learning and memory ability were significantly lower than the blank control group. Compared with the model control group, the 24h memory capacity and AchE content were not significant. Sexual differences. Conclusion: In the combination therapy of Yiqi Huoxue for the treatment of ischemic cerebrovascular diseases, the dosage of Astragalus should be large, but as the dose increases, the brain protection effect is weakened. Its dosage is about 60g/d.
