TNT是存在于肌肉组织的一种调节蛋白,具有结合原肌球蛋白的功能。心肌TNT的表达特异,因此,骨骼肌和心肌TNT异构体蛋白结构差异显著,用免疫学方法不难将心肌与骨骼肌的TNT分开,TNT测定在心脏缺血性疾病诊断中有较大价值。通过盐析、阴离子交换交换层析及聚焦色谱法(CF)得到纯度较高TNT,经SDS-PAGE、等电聚焦电泳(IEF)分析为一条蛋白区带,等电点为7.35,分子量37.4KD。纯化心肌TNT,为研究TNT特性,建立检测TNT方法提供了必要条件。
TNT is a regulatory protein present in muscle tissue that binds tropomyosin. Therefore, there are significant differences in the structure of TNT isoforms between skeletal muscle and myocardium. It is not difficult to separate the myocardial and skeletal muscle TNT by immunological methods. TNT has great value in the diagnosis of cardiac ischemic disease . High purity TNT was obtained by salting-out, anion exchange chromatography and focused chromatography (CF), and was analyzed by SDS-PAGE and isoelectric focusing (IEF) as a protein band with an isoelectric point of 7.35 and a molecular weight 37.4KD. Purification of myocardial TNT provides a necessary condition for the study of TNT properties and establishment of a method for the detection of TNT.
