为了提高血吸虫病病因诊断的特异性和敏感性,用慢性日本血吸虫病病人血清筛选以λgtll构建的日本血吸虫中国大陆株成虫cDNA表达库,获得一阳性克隆Sjgt4,其表达产物为148000融合蛋白。插入的cDNA片段经聚合酶链反应(PCR)法扩增、纯化后,亚克隆入高表达载体pGEX,获得克隆SjpG5,其表达产物为58000融合蛋白,应用免疫印迹技术,其可被96%(24/25)的日本血吸虫病病人血清中特异抗体所识别。
In order to improve the specificity and sensitivity of the diagnosis of schistosomiasis, the cDNA library of adult Schistosoma japonicum strains with λgtll was screened from serum of patients with chronic schistosomiasis japonica, and a positive clone Sjgt4 was obtained. The expressed product was 148000 fusion protein. The inserted cDNA fragment was amplified by polymerase chain reaction (PCR), purified and subcloned into high expression vector pGEX to obtain clone SjpG5 whose expression product was 58000 fusion protein. 24/25) of schistosomiasis japonica patients serum specific antibodies recognized.
