目的:探讨地塞米松对~(125)I放射性粒子内照射A549肺癌细胞AQP5及存活素(survivin)表达的影响。方法:共25只A549肺腺癌皮下种植BALB/c-nu裸鼠模型随机分为实验组(n=12)和对照组(n=13),实验组进行~(125)I放射性粒子植入+地塞米松(鼠尾静脉注射),对照组进行~(125)I放射性粒子植入,7 d后处死裸鼠行AQP5及survivin免疫组化检测。结果:实验组的AQP5阳性表达率高于对照组(分别为91.6%和53.8%,P=0.046),survivin的阳性表达率低于对照组(分别为33.3%和76.9%,P=0.036)。结论:地塞米松可能通过上调AQP5的表达进而增加A549肺癌细胞对~(125)I的辐射敏感性,其机制与survivin表达抑制相关。
Objective: To investigate the effect of dexamethasone on the expression of AQP5 and survivin in A549 lung cancer cells irradiated with ~ (125) I radioactive particles. Methods: A total of 25 A549 lung adenocarcinoma subcutaneously BALB / c nude mice were randomly divided into experimental group (n = 12) and control group (n = 13). The experimental group received 125 I radioactive seed implantation + Dexamethasone (tail vein injection). The control group received ~ (125) I radioactive seed implantation. AQP5 and survivin immunohistochemistry were performed on the 7th day after the mice were sacrificed. Results: The positive rate of AQP5 expression in the experimental group was higher than that in the control group (91.6% and 53.8%, P = 0.046, respectively). The positive rate of survivin in the experimental group was lower than that in the control group (33.3% and 76.9%, P = 0.036). Conclusion: Dexamethasone may increase the sensitivity of ~ (125) I to A549 lung cancer cells by up-regulating the expression of AQP5. The mechanism is related to the inhibition of survivin expression.
