应用毛细管区带电泳-紫外检测器(CZE-UV)法同时分离测定猪肉中4种核苷酸。采用阴极检测方法,考察了硼砂浓度、运行缓冲液浓度及p H值、分离电压、毛细管温度4个因素对分离效果和重现性的影响。试验得出最优分离条件为:分离电压25 k V、检测波长214 nm、毛细管温度25℃,以12 mmol/L的硼砂(p H10.0)为运行缓冲液,4种核苷酸在13 min内达到基线分离;检出限最低可达0.4 mg/L;线性范围为(20~100)mg/L;相关系数(R~2)均大于0.997;日内精密度为0.37%~3.97%;日间精密度为1.29%~10.8%。实际猪肉样品中4种核苷酸的加标回收率为103.69%~107.09%。结果表明CZE-UV方法简单、快速、实用,可为猪肉质量评估提供借鉴方法。
Simultaneous Determination of Four Nucleotides in Pork by Capillary Zone Electrophoresis - UV Detector (CZE-UV). The effects of borax concentration, running buffer concentration and p H value, separation voltage and capillary temperature on separation efficiency and reproducibility were examined by cathodic detection method. The optimum separation conditions were as follows: separation voltage 25 kV, detection wavelength 214 nm, capillary temperature 25 ℃, 12 mmol / L borax (p H10.0) as running buffer and four kinds of nucleotides at 13 The linear range was (20-100) mg / L; the correlation coefficient (R ~ 2) was greater than 0.997; the intra-day precision was 0.37% -3.97%; the detection limit was as low as 0.4 mg / Precision during the day was 1.29% ~ 10.8%. The spiked recoveries of four nucleotides in real pork samples ranged from 103.69% to 107.09%. The results show that the CZE-UV method is simple, rapid and practical, which can provide a reference for the quality evaluation of pork.
