目的 :筛选对视网膜组织细胞具有药物保护作用的中药成分。方法 :体外细胞培养 ,新生小牛视网膜细胞悬液为研究对象 ,SFA ,苦参碱 ,青藤碱 ,紫草素为处理因素 ,维拉帕米和神经生长因子 (NGF)为处理对照 ,行细胞培养 2 4小时后MTT法检测细胞增殖活性。结果 :SFA对视网膜组织细胞有明显的促增殖作用 ,且呈剂量依赖关系 ;苦参碱轻度促进视网膜组织细胞增殖 ;青藤碱 ,维拉帕米和NGF轻度抑制视网膜组织细胞增殖 ;而紫草素则低浓度时抑制 ,高浓度时促进视网膜组织细胞增殖。结论 :SFA ,苦参碱对视网膜组织细胞具有较好的药物保护作用
Objective : To screen Chinese medicinal ingredients with protective effects on retinal tissue cells. METHODS: In vitro cell culture, retinal cell suspensions of newborn calves were studied, SFA, matrine, sinomenine, shikonin as the treatment factors, verapamil and nerve growth factor (NGF) as treatment controls. After 24 hours of cell culture, cell proliferation activity was measured by MTT assay. RESULTS: SFA had a significant proliferative effect on retinal tissue cells in a dose-dependent manner. Matrine slightly promoted cell proliferation in retinal tissues. Sinomenine, verapamil and NGF slightly inhibited cell proliferation in retinal tissues; Shikonin is inhibited at low concentrations and promotes cell proliferation in retinal tissues at high concentrations. Conclusion : SFA and matrine have better drug protection on retinal tissue cells
