蚯蚓纤溶酶对溶栓,纤溶和抗凝作用的影响

实验表明蚯蚓纤溶酶不仅具有体外溶解血栓和血块的作用,口服300万mm~2/kg和静注10万mm~2/kg也可加速急性肺动脉栓塞的溶栓效应,与生理盐水组比有显著性差异。纤维蛋白平板试验,点样后3.5h蚯蚓纤溶酶3000mm~2/ml与上海生化制药厂生产的尿激酶250u/ml的纤溶活力相似。此外,它能使兔血浆纤维蛋白原含量下降,“三P”试验阳性,兔凝血时间,复钙时间和凝血酶原时间延长。我国血栓栓塞性疾病的发病率同日本,欧美一样逐年增加,成为严重危害人民身体健康的心血管疾病。因此需要尽快寻找一种疗效好,不良反应少,应用方便和价廉的溶栓药物。近几年来国内外对蚯蚓的抗栓作用虽进行了一些研究,但未见动物体内溶栓试验报道。本文应用兰州大学研制的,山东济南生化制药厂生产的蚯蚓纤溶酶,进行了溶栓,纤溶和抗凝方面的临床前药理研究,为临床试验提供实验依据。 Experiments show that earthworm fibrinolytic not only has the function of dissolving thrombus and blood clot in vitro, but oral administration of 3 million mm ~ 2 / kg and intravenous injection of 100,000 mm ~ 2 / kg can also accelerate the thrombolysis effect of acute pulmonary embolism, compared with saline group There are significant differences. In recent years, the research on the antithrombotic effects of earthworms both at home and abroad has been done, but no reports of thrombolysis in animals have been reported. In this paper, the earthworm fibrinolytic enzyme produced by Lanzhou University and produced by Jinan Biochemical Pharmaceutical Factory in Shandong Province was used to carry out preclinical pharmacological studies on thrombolysis, fibrinolysis and anticoagulation to provide experimental evidence for clinical trials.
