目的探讨Shc与大肠癌TNM分期、分化及转移的相关性。方法选取2010年1月至2013年10月齐齐哈尔医学院附属第三医院收治的资料完整的经病理学确诊的大肠癌标本79例,随机取其中20例距癌组织5 cm外的正常组织,即癌旁组织为对照。免疫组化法检测大肠癌和癌旁组织的Shc表达,Western Blot定量大肠癌组织p52Shc、p46Shc和p66Shc表达,组间比较采用t检验。结果相比于癌旁组织,Shc在大肠癌组织中呈现高表达,在大肠癌细胞核及细胞质中均有Shc蛋白的表达增强,细胞质中显著。与p66Shc比,大肠癌组织的p52Shc和p46Shc表达较强。大肠癌分化程度越低(p52Shc:2.9±0.3 vs.3.1±0.5 vs 3.4±0.4,p46Shc:2.1±0.3 vs 2.3±0.4 vs 2.6±0.6)、TNM分期越高(p52Shc:0.0±0.0 vs 3.0±0.2 vs 3.2±0.5 vs 3.6±0.7,p46Shc:0.0±0.0 vs 2.2±0.2 vs 2.6±0.4 vs 2.7±0.6)、浸润越深(p52Shc:0.0±0.0 vs 2.8±0.4 vs 3.0±0.4 vs 3.3±0.2,p46Shc:0.0±0.0 vs 2.0±0.3 vs 2.3±0.3 vs 2.5±0.3)、远处转移(p52Shc:3.5±0.5 vs 3.1±0.6,p46Shc:2.6±0.2 vs 2.4±0.3)和淋巴结转移(p52Shc:3.0±0.5 vs 3.4±0.4,p46Shc:2.5±0.4 vs 2.3±0.2)的p52Shc和p46Shc表达明显增高(P<0.05或P<0.01)。结论 Shc的高表达与大肠癌的发生发展可能存在联系,尤其p52Shc、p46Shc与大肠癌的发生发展关系密切。
Objective To investigate the correlation between Shc and TNM staging, differentiation and metastasis of colorectal cancer. Methods From January 2010 to October 2013, Qiqihar Medical College Affiliated Third Hospital admitted to the data of 79 cases of pathologically confirmed colorectal cancer specimens were randomly selected from 20 cases of cancer tissue 5 cm away from the normal tissue, that is, Paracancerous tissue as a control. Immunohistochemical method was used to detect the expression of Shc in colorectal cancer and paracancerous tissues. Western Blot was used to quantify the expression of p52Shc, p46Shc and p66Shc. The t test was used to compare the two groups. Results Compared with the adjacent tissues, Shc was highly expressed in colorectal cancer tissues. The expression of Shc protein was enhanced in the nuclei and cytoplasm of colorectal cancer, and the expression of Shc was significant in the cytoplasm. Compared with p66Shc, p52Shc and p46Shc were strongly expressed in colorectal cancer. The lower the degree of colorectal cancer differentiation was (p52Shc: 0.0 ± 0.0 vs. 3.0 ± 0.4 vs 0.3 ± 0.4 vs 0.4 ± 0.4, p46Shc: 2.1 ± 0.3 vs 2.3 ± 0.4 vs 2.6 ± 0.6) 0.2 vs 3.2 ± 0.5 vs 3.6 ± 0.7, p46Shc: 0.0 ± 0.0 vs 2.2 ± 0.2 vs. 2.6 ± 0.4 vs 2.7 ± 0.6), with deeper infiltration (p52Shc: 0.0 ± 0.0 vs 2.8 ± 0.4 vs 3.0 ± 0.4 vs. 3.3 ± 0.2 , p46Shc: 0.0 ± 0.0 vs 2.0 ± 0.3 vs. 2.3 ± 0.3 vs. 2.5 ± 0.3), distant metastasis (p52Shc: 3.5 ± 0.5 vs 3.1 ± 0.6, p46Shc: 2.6 ± 0.2 vs. 2.4 ± 0.3), and lymph node metastasis (p52Shc: 3.0 ± 0.5 vs 3.4 ± 0.4, p46Shc: 2.5 ± 0.4 vs 2.3 ± 0.2), the expressions of p52Shc and p46Shc were significantly increased (P <0.05 or P <0.01). Conclusion The high expression of Shc and the occurrence and development of colorectal cancer may be related, especially p52Shc, p46Shc and the occurrence and development of colorectal cancer are closely related.
