目的研究不同培养条件对铁皮石斛类原球茎在生物反应器培养的影响。方法采用接种量、通气量、蔗糖浓度、光照强度等单因素试验设计,烘干法测定生物量,苯酚-硫酸法测定多糖量,数据采用SPSS 16.0软件分析。结果在生物反应器培养条件下,利于铁皮石斛类原球茎增殖和多糖积累的培养基组分为1/2 MS基本培养基添加1.0 mg/L NAA、5%椰乳、3%蔗糖,pH值为6.0;接种量为40 g/L,采用孔径为15μm的多孔喷头,通气量用1.0 L/min和0.5 L/min交替使用以及光照强度为2 000 lx等培养条件有效地提高3类原球茎增殖系数和多糖量。结论不同的培养条件对铁皮石斛类原球茎的生长、多糖的变化有显著影响,适宜的培养条件对铁皮石斛类原球茎的生物量和主要药用成分的生产具有重要的实践意义。
Objective To study the effects of different culture conditions on the culture of Dendrobium officinale protocorm in bioreactor. Methods The single factor experiments such as inoculum size, aeration rate, sucrose concentration and light intensity were used to determine the biomass. The biomass was determined by drying method. The polysaccharide content was determined by phenol - sulfuric acid method. The data were analyzed by SPSS 16.0 software. Results Under the condition of bioreactor culture, the components of the medium that favored the proliferation and polysaccharide accumulation of P. officinale protocorms were 1.0 mg / L NAA, 5% coconut milk, 3% sucrose, pH value Was 6.0; inoculum size was 40 g / L, and the number of protoplasts was increased by using multi-hole nozzle with 15 μm pore size. The aeration volume was alternately increased by 1.0 L / min and 0.5 L / min and the illumination intensity was 2 000 lx. Proliferation coefficient and polysaccharide content. Conclusion Different culture conditions have significant effects on the growth of the protocorm-like bodies of Dendrobium candidum and the changes of polysaccharides. Appropriate culture conditions have important practical significance for the biomass of the protocorm-like bodies of Dendrobium candidum and the production of the main medicinal components.
