目的:探讨Vav3及雌激素受体(ER)在乳腺癌组织中的表达情况及临床意义。方法:免疫印迹法检测乳腺癌细胞及正常乳腺上皮细胞中Vav3的表达;SP免疫组化法检测45例乳腺癌、10例癌旁正常乳腺组织中Vav3及ER的表达情况。结果:乳腺癌细胞(MCF-7)中Vav3蛋白表达为1.659±0.106,正常乳腺上皮细胞(MCF-10A)中Vav3蛋白表达为0.182±0.692,两者比较,差异有统计学意义(P<0.05)。乳腺癌组织中Vav3蛋白阳性表达率为87.5%,癌旁正常乳腺组织中Vav3蛋白阳性表达率为0,两者比较,差异有统计学意义(P<0.05)。Vav3阳性表达与ER阳性表达呈显著正相关(r=6.334,P=0.012)。结论:Vav3的表达与乳腺癌的发生发展有关,并且可通过激活ER信号通路调控肿瘤生长。
Objective: To investigate the expression and clinical significance of Vav3 and estrogen receptor (ER) in breast cancer. Methods: The expression of Vav3 in breast cancer cells and normal breast epithelial cells was detected by Western blotting. The expression of Vav3 and ER in 45 cases of breast cancer and 10 cases of adjacent normal breast tissues were detected by SP immunohistochemistry. Results: The expression of Vav3 protein in breast cancer cells (MCF-7) was 1.659 ± 0.106 and that in normal mammary epithelial cells (MCF-10A) was 0.182 ± 0.692, the difference was statistically significant (P <0.05 ). The positive expression rate of Vav3 protein in breast cancer tissues was 87.5%, while the positive expression rate of Vav3 protein in adjacent normal breast tissues was 0. The difference was statistically significant (P <0.05). Vav3 positive expression was positively correlated with ER positive expression (r = 6.334, P = 0.012). Conclusion: The expression of Vav3 is related to the occurrence and development of breast cancer, and it can regulate tumor growth by activating ER signaling pathway.
