对苏丹草进行甘蔗条纹花叶病毒(SCSMV)人工接种,其叶片呈现连续褪绿条斑症状。采用RT-PCR和SC-SMV特异性引物SCSMV-ST2(F)、SCSMV-P1(R)对苏丹草染病植株上的甘蔗条纹花叶病毒进行鉴定,结果为阳性;对所获得的PCR扩增产物进行克隆和序列分析,获得0.5kbp片段,在GenBank数据库中的注册号为No.EU650178;通过BLAST比对和系统遗传分析(MEGA 4.0),将其鉴定为甘蔗条纹花叶病毒,这是首次发现苏丹草可作为一种甘蔗条纹花叶病毒的人工选择寄主。
Artificial inoculation of Sudan grass with sugarcane stripe mosaic virus (SCSMV) showed continuous leaf streaking symptoms. SCSMV-ST2 (F) and SCSMV-P1 (R) were used for identification of sugarcane streak mosaic virus on Sudan grass infected plants with RT-PCR and SC-SMV primers. The results were positive. The product was cloned and sequenced to obtain a 0.5kbp fragment with the accession number No. EU650178 in the GenBank database. This was identified as sugarcane streak mosaic virus by BLAST alignment and phylogenetic analysis (MEGA 4.0) Sudan was found to be an artificial selection host for sugarcane stripe mosaic virus.
