目的为完善小鼠早期胚胎体外培养体系,了解槲皮素对小鼠1-细胞胚胎体外发育的影响。方法以CZB为基础培养液,添加不同浓度(500、50、5、1、0.5、0.2、0.1μm)的槲皮素作为试验组,添加等量二甲基亚砜(DMSO)作为对照组,观察不同浓度槲皮素对胚胎发育到囊胚各阶段的影响。结果当槲皮素浓度超过50μm时,对胚胎发育有毒性效应;0.1~1μm浓度槲皮素对胚胎发育有促进作用,特别是0.5μm槲皮素能明显提高胚胎的囊胚形成率。结论 0.5μm槲皮素能促进小鼠早期胚胎的体外发育。
The aim is to improve the in vitro culture system of mouse early embryos and to understand the effect of quercetin on the in vitro development of mouse 1-cell embryos. Methods CZB-based culture medium was added with different concentration of quercetin (500,50,5,1,0.5,0.2,0.1μm) as the experimental group, the same amount of dimethylsulfoxide (DMSO) as control group, The effects of different concentrations of quercetin on the stages of embryo development to blastocysts were observed. Results When the concentration of quercetin was more than 50μm, it had toxic effect on embryo development. Quercetin 0.1μm ~ 1μm could promote embryo development, especially 0.5μm quercetin could significantly improve blastocyst formation rate. Conclusion 0.5μm quercetin can promote the development of mouse embryos in vitro.
