目的 探讨硫芥诱导体外培养大鼠脾脏淋巴细胞凋亡及与细胞周期的关系。方法 体外分离培养大鼠脾脏淋巴细胞。以流式细胞仪、琼脂糖凝胶电泳分别检测细胞凋亡 ,细胞周期 ,和DNA片段改变。结果 硫芥可诱导淋巴细胞凋亡 (P <0 0 5 ) ,对细胞周期有明显影响 ,主要是S期和G2 /M期明显降低 ,引起淋巴细胞生长阻滞在G1期 ;流式细胞仪PI染色出现亚二倍体峰 (凋亡峰 ) ,其G1期DNA含量明显增加 ,S期含量减少 ;琼脂糖凝胶电泳呈典型的“梯状”带 (DNAladder)。结论 硫芥可抑制体外培养的淋巴细胞由G1期进入S期 ,抑制体外培养的淋巴细胞增殖 ,通过诱导淋巴细胞凋亡实现作用机制。
Objective To investigate the apoptosis of spleen lymphocytes induced by sulfur mustard in vitro and its relationship with cell cycle. Methods Rat spleen lymphocytes were isolated and cultured in vitro. Flow cytometry and agarose gel electrophoresis were used to detect apoptosis, cell cycle, and DNA fragment changes. Results Sulfur mustard induced lymphocyte apoptosis (P <0 05), and had a significant effect on cell cycle, mainly S phase and G2 / M phase was significantly reduced, causing lymphocyte growth arrest in G1 phase; flow cytometry PI staining showed sub-diploid peak (apoptotic peak), the DNA content of G1 phase increased significantly, S phase decreased; agarose gel electrophoresis showed a typical “ladder” (DNAladder). Conclusion Sulfur mustard can inhibit the proliferation of lymphocytes cultured in vitro from the G1 phase to the S phase and inhibit the proliferation of lymphocytes cultured in vitro and induce the apoptosis of lymphocytes.
