目的探讨慢性坐骨神经嵌压伤(CCI)模型小鼠背根神经节(DRG)电压门控性钙通道3.2(Cav3.2)变化对早期生长反应蛋白2(Egr2)表达的影响。方法 80只C57BL/6小鼠随机分为四组:阴性对照(A)组、CCI模型(B)组、米贝地尔干预(C)组和细胞外调节蛋白激酶(ERK)抑制剂U0126干预(D)组。B、C、D组行坐骨神经嵌压手术建立CCI模型。行为学检测机械痛觉和热痛觉,Western blot测定DRG中Cav3.2、总ERK、磷酸化ERK(pERK)和Egr2蛋白水平,RT-PCR测定DRG中Egr2基因水平。结果与A组相比,B组手术后出现机械痛觉超敏和热痛觉过敏(P<0.05)。C、D组机械痛觉和热痛觉均较B组不同程度缓解(P<0.05)。Cav3.2、pERK和Egr2蛋白表达呈时间依赖改变。与A组相比,C组pERK蛋白水平下降(P<0.05),C、D组Egr2蛋白和基因水平下降(P<0.05)。结论在CCI模型小鼠的慢性坐骨神经损伤中,ERK信号通路可能是Cav3.2钙通道调控Egr2蛋白和基因表达的重要途径之一。
Objective To investigate the effect of voltage-gated calcium channel 3.2 (Cav3.2) in dorsal root ganglion (DRG) on the expression of early growth factor 2 (Egr2) in chronic sciatic nerve crush injury (CCI) model mice. Methods Eighty C57BL / 6 mice were randomly divided into four groups: negative control group (A), CCI model group (B), mibefil intervention group (C) and extracellular regulated protein kinase (ERK) inhibitor U0126 intervention (D) group. The CCI model was established in the B, C and D groups by sciatic nerve entrapment. Behavioral tests were performed to detect mechanical pain and thermal pain. The levels of Cav3.2, total ERK, phosphorylated ERK (pERK) and Egr2 in DRG were detected by Western blot. The level of Egr2 gene in DRG was determined by RT-PCR. Results Compared with group A, mechanical allodynia and thermal hyperalgesia occurred after operation in group B (P <0.05). The mechanical pain and thermal pain in groups C and D were relieved to some extent compared with those in group B (P <0.05). Cav3.2, pERK and Egr2 protein expression in a time-dependent manner. Compared with group A, the level of pERK protein in group C decreased (P <0.05), while the level of Egr2 protein and gene in group C and D decreased (P <0.05). Conclusion ERK signaling pathway may play an important role in the regulation of Egr2 protein and gene expression by Cav3.2 calcium channel in chronic sciatic nerve injury of CCI model mice.
