目的测定人巨噬细胞金属弹性蛋白酶(HME)在胃癌细胞及胃癌组织中的表达,评价其在胃癌进展中的作用。方法采用实时荧光PCR测定胃癌细胞及胃癌组织中HME mRNA表达水平,并对胃癌组织中HME mRNA表达与患者临床病理特征及预后的关系进行统计分析。结果MGC-803、SGC-7901和AGS胃癌细胞株均有HME mRNA的表达。胃癌组织、癌旁组织和正常组织的HME mRNA表达阳性率分别为31.03%、25.87%和18.97%,阳性结果拷贝数对数值分别为6.06±0.76、3.53±0.85和1.88±0.82,胃癌组织HME tuRNA表达阳性率、HME mRNA表达量均显著高于正常组织(P<0.05)。HME mRNA表达与肿瘤的位置、大小、浸润深度、耐药基因topⅡ和PG的表达无关,胃癌组织HME mRNA阳性患者淋巴结转移发生率低于阴性者、GST基因表达阳性率低于阴性者、2年生存率高于阴性者。结论胃癌组织中HME mRNA表达阳性与胃癌转移潜能降低有关,可能是预后良好的标志。
Objective To determine the expression of human macrophage metalloelastase (HME) in gastric cancer cells and gastric cancer and to evaluate its role in the progression of gastric cancer. Methods The expression of HME mRNA in gastric cancer cells and gastric cancer tissues was detected by real-time fluorescence PCR. The relationship between the expression of HME mRNA in gastric cancer tissues and clinicopathological characteristics and prognosis was analyzed. Results The expression of HME mRNA in both MGC-803, SGC-7901 and AGS gastric cancer cell lines. The positive rates of HME mRNA expression in gastric cancer tissue, paracancerous tissue and normal tissue were 31.03%, 25.87% and 18.97%, respectively. The logarithmic copy number of positive results were 6.06 ± 0.76, 3.53 ± 0.85 and 1.88 ± 0.82, respectively. HME tuRNA The positive expression rate of HME mRNA was significantly higher than that of normal tissues (P <0.05). The expression of HME mRNA was not related to the tumor location, size, depth of invasion, topⅡ and PG expression, but the incidence of lymph node metastasis in HME mRNA positive patients was lower than that in negative ones. The positive rate of GST expression in gastric cancer tissues was lower than that in negative ones Survival rate is higher than negative. Conclusion The positive expression of HME mRNA in gastric cancer is related to the decreased metastatic potential of gastric cancer, which may be a good marker of prognosis.
