目的比较石蜡组织芯片和相应常规组织切片正常成人肺泡Ⅱ型上皮细胞、人胚胎肺泡上皮细胞和肺癌细胞的细胞核体视学参数,探讨组织芯片代表性的可靠程度。方法收集肺癌手术切除标本:鳞癌52例、腺癌38例、大细胞癌10例、小细胞癌4例、正常肺组织20例和胚胎肺组织20例。采用组织微阵列技术构建760点阵石蜡组织芯片。用Image-Pro图像分析软件测试胞核及细胞,按体视学公式计算以细胞为参照空间时肺癌细胞核的体积密度(Vvn)、表面积密度(Svn)、表面积与体积比(Rsv,n)、核浆比(Rnp)、平均自由程(λn)。结果石蜡组织芯片和相应常规组织切片细胞核的体视学参数基本类同,两者比较差异无显著性,P值均>0.05。结论组织芯片和相应常规组织切片细胞核的体视学参数基本类同,组织芯片具有较好的可靠性。
Objective To compare the nuclear visual parameters of normal adult alveolar type Ⅱ epithelial cells, human embryonic alveolar epithelial cells and lung cancer cells from paraffin wax tissue and corresponding routine tissue sections and to discuss the reliability of representativeness of tissue microarray. Methods Surgical specimens of lung cancer were collected from 52 cases of squamous cell carcinoma, 38 cases of adenocarcinoma, 10 cases of large cell carcinoma, 4 cases of small cell carcinoma, 20 cases of normal lung tissue and 20 cases of embryonic lung tissue. Tissue microarray technique was used to construct a 760-dot matrix paraffin tissue chip. The nuclei and cells were tested by Image-Pro image analysis software. The Vvn, Svn, Rsv, n of lung cancer cell nucleus were calculated according to the stereological formula. Nuclear ratio (Rnp), mean free path (λn). Results There was no significant difference in the stereological parameters between the paraffin-embedded tissue sections and the corresponding routine tissue section nuclei. The P values were both> 0.05. Conclusion Tissue chips and the corresponding routine tissue sections nuclei stereological parameters are basically the same, the tissue chip has good reliability.
