目的 :深入了解RB基因过表达在大肠癌发生、发展中的意义。方法 :利用Northernblot,Dotblot在 1 6例大肠癌及 1 5例大肠腺瘤中测定RB基因的表达情况。运用核酸酶保护试验观察RB基因的点突变现象。结果 :发现在大多数大肠癌 ( 1 4/1 6 )及部分大肠腺瘤 ( 2 / 1 5 )中 ,同相应的正常大肠粘膜相比 ,存在着RB基因过表达现象。在 2例RB基因过表达的大肠癌标本中观察到RB基因发生了点突变。结论 :导致突变失活型Rb蛋白含量增高的RB基因异常改变与大肠癌发生、发展可能密切相关
Objective : To further understand the significance of RB gene overexpression in the development and progression of colorectal cancer. Methods : The expression of RB gene was detected in 16 cases of colorectal cancer and 15 cases of colorectal adenoma using Northern blot and Dotblot. Nuclease protection assay was used to observe the point mutation of RB gene. RESULTS: Overexpression of RB gene was observed in most colorectal cancers (14/16) and some colorectal adenomas (2/15) compared with the corresponding normal colorectal mucosa. A point mutation was observed in the RB gene in two cases of colorectal carcinoma overexpressing RB gene. Conclusion :Abnormal changes of RB gene that lead to the increase of mutant inactive Rb protein may be closely related to the occurrence and development of colorectal cancer
