目的 建立一种简单、经济的血小板衍化生长因子(PDGF)的纯化方法 .方法 血小板衍化生长因子 (PDGF)具有耐酸性 (p H 2 .5 ) ,耐热 (10 0℃ )以及耐受 2 % SDS的特性 .采用酸醇提取、热处理、离子交换层析、分子筛等蛋白分离和纯化技术从猪血小板中提取和纯化 PDGF.结果 猪血小板衍化生长因子 (p PDGF)纯化倍数达 75 82倍 ,活性回收率为3% .结论 本方法简单、经济 ,纯化的猪血小板衍化生长因子(p PDGF)具有明显的生物学活性
Objective To establish a simple and economical method for purification of platelet-derived growth factor (PDGF) .Methods Platelet-derived growth factor (PDGF) has the properties of acid resistance (p H 2 .5), heat tolerance (100 ℃) SDS was used to extract and purify PDGF from porcine platelets by acid-alcohol extraction, heat treatment, ion-exchange chromatography, molecular sieve and other protein isolation and purification methods.Results The purification of porcine PDGF was 75.82 fold, and the activity The recovery rate was 3% .Conclusion The method is simple, economical and purified porcine platelet-derived growth factor (p PDGF) has obvious biological activity
